I put new speakers into the front doors yesterday but due to the odd speaker opening shape, there is an open area around the speaker. Will this make any difference in the sound quality having the open area between the outter edge of the speaker and the speaker opening in the door?

not really.. .but you should have made a plate out of 1/8" MDF to make up for the 'odd speaker opening'
